I've tried USB 2.0 1GB JumpDrive, USB 2.0 CF Reader, & USB 2.0 80G External HD and none of them were recognized by the Xbox 360.
I've not tried converting it to NTFS...
I'll try that later today and will let you know..

I was really hoping that I could just hookup my external USB HD and play my 200GB collection of WMA files.

I hooked up my 1GB SanDisk Cruzer Mini thumb drive and it worked perfectly. (It's FAT btw... so that may not help you, lol) I've yet to try my external HDD though... I'll try it tomorrow and let you know how that goes as well. But it too is FAT32, but I'll let ya know anyways.

Just incase some people are about to waste some time....
I have a usb hub on the controller for my xbox1, and I've used a 256mb flash drive to move saves to friends houses just by bringing the controller over and using the flash drive.
I've freshly formatted the flash drive through the x1 dashboard to approximate a xbox1 mem card and the 360 will not reconize it atall, beter off to leave it in fat32 and you'll be able to atleast play tracks off it.

Yes it will work but the xbox 360 only reads image files (.jpg exc) and music files (.mp3 exc)

Llama was able to hookup a 60GB USB HD to their 360.

NTFS doesn't seem to work, so yeah, leave it FAT32 or FAT. It'll show up under portable devices (? I think that's it), not under memory.
