Yes, You only need run the .iso (assuming you still have it) thru AGBX and then re-burn.

My guess would be that ur original had either dust or a scratch before you ripped it (although you never know, without knowing ur region it's difficult to tell but usually if it doesn't pass the initial check then it won't boot period), Make sure you verify after burning, as verification will confirm that the problem is with the image on ur hdd (AGBX wont pick-up most basic forms of data corruption) ie; If ImgBurn/XBC verifys after writing then don't bother burning it again (let alone 5 times) Re-Rip and Re-Patch
I suspect that the new dash is carrying-out checks though at other intervals eg; when you go back to dash but it's unlikely that ur issue has anything to do with AP 2.5
